when is the best time to count cards?

Personally, I like to count in the morning, when I am still fresh. I think it is a bad idea to play when you are tired. It can also be good to go at off peak times, when it is easier to find a heads up game. Other patrons at the casino can be sooooo annoying at the blackjack table, when they agonize over the most trivial decisions or get bellicose because of the way you play. Playing heads up is a lot more profitable too, because you can get in so many more rounds per hour.

That's true. A busy night will keep the pit man running around, with fills, payouts to verify, and all the other minutiae of their job, whereas if it is deserted they will almost have to focus on you, if only out of boredom. Some times just before the evening rush is a good time to hit, when they open a bunch of tables but the patrons haven't quite started to roll in yet. Or at the end of the night, when most of the ploppies have gone home, and the dealers might be a little more error prone, on account of their fatigue. I guess this can vary from store to store as well.
